Cyclomics launches CySeq®: a solution for PCR-Free, Strand-Resolved and Ultra-Accurate Cell-Free DNA Sequencing

Cyclomics, a pioneer in precision diagnostics solutions, today announces the commercial launch of CySeq®, a breakthrough product designed for highly accurate cell-free DNA (cfDNA) analysis. CySeq® addresses fundamental limitations of existing cfDNA sequencing approaches, opening new possibilities for liquid biopsy analysis, oncology research, non invasive prenatal testing, and the emerging field of ctDNA fragmentomics.


Cell-free DNA circulates in blood at low concentrations, making it notoriously difficult to identify rare cancer mutations from cfDNA. CySeq® technology concatenates each cfDNA molecule, and the resulting concatemers can be analyzed by any sequencer capable of generating sequencing reads above ~1000bp. The accompanying software suite generates highly-accurate consensus sequences. Cancer mutations can thus be identified at single molecule level.


Unlike conventional approaches that capture only one form of cfDNA, CySeq® captures both single-stranded and double-stranded cfDNA molecules within a single workflow. This comprehensive capture ensures that no biologically and clinically meaningful fraction of the cfDNA landscape is overlooked — a critical advantage for measuring the presence of cancer from cfDNA.


Perhaps most significantly, CySeq® is the first cfDNA sequencing product to link the forward and reverse strands of the same double-stranded cfDNA molecule, without any modification of cfDNA ends, thus providing a complete and accurate picture of each individual DNA fragment.


This strand-resolved approach unlocks a perfect view on fragmentomics — the study of cfDNA fragment length, end motifs, and positional information that encodes rich biological signals about tissue of origin, chromatin organisation, and nucleosome positioning. Researchers and clinicians can now access this layer of information with confidence, without artefacts due to standard DNA library preparations, which confound fragment-level analyses.

The solution works well with any long-read sequencing platform, and has been particularly tested in conjunction with Oxford Nanopore Sequencing. A version of CySeq® for use with PCR amplicons is also available, enabling custom or existing PCR panels to be sequenced at ultra-high accuracy due to the inherent CySeq® consensus sequencing capabilities.
